Currently, in vitro embryo production (IVP) is successfully commercially applied in cattle. However, the high sensitivity of embryos to cryopreservation in comparison to in vivo (IVD) embryos slows the dissemination of this biotechnology. Reduced cryotolerance is frequently associated with lipid accumulation in the cytoplasm mainly due to in vitro culture conditions. The objective of this study was to evaluate the lipid composition of biopsied and sexed embryos, produced either in vivo or in vitro from the same Holstein heifers before and after a slow freezing protocol. Lipid extracts were analysed by liquid chromatography-high resolution mass spectrometry, which enabled the detection of 496 features. Our results highlighted a lipid enrichment of IVP embryos in triglycerides and oxidised glycerophospholipids and a reduced abundance in glycerophospholipids. The slow freezing process affected the lipid profiles of IVP and IVD embryos similarly. Lysophosphatidylcholine content was reduced when embryos were frozen/thawed. In conclusion, the embryonic lipid profile is impacted by IVP and slow freezing protocols but not by sex. Lysophosphatidylcholine seemed highly sensitive to cryopreservation and might contribute to explain the lower quality of frozen embryos. Further studies are required to improve embryo freezability by modulating the lipidome.Two special cases of dart leader propagation were observed by the high-speed camera in the leader/return stroke sequences of a classical triggered lightning flash and an altitude-triggered lightning flash, respectively. Different from most of the subsequent return strokes preceded by only one leader, the return stroke in each case was preceded by two leaders occurring successively and competing in the same channel, which herein is named leader-chasing behavior. In one case, the polarity of the latter leader was opposite to that of the former leader and these two combined together to form a new leader, which shared the same polarity with the former leader. The present study aimed to determine the contribution of quadriceps strength, power and endurance to daily PA in COPD. Additionally, for quadriceps endurance, we also aimed to determine to what extent the association varies according to the mode of movement (isotonic, isometric, or isokinetic). Using a multicentre cross-sectional trial design we determined the contribution of quadriceps function to daily PA (steps, sedentary time and time spent doing moderate-to-very-vigorous physical activity [MVPA]) using bivariate and partial Pearson correlation analysis (r) and multiple linear regression models (ΔR2). Pre-determined controlling factors were sex, age, body mass index (BMI), COPD-assessment test, forced expiratory volume in one second in percent of the predicted value (FEV1pred), and distance walked on the 6-minute walk test. Eighty-one patients with COPD (mean ± SD age 67 ± 8 years, FEV1pred 57 ± 19%, daily steps 4968 ± 3319, daily sedentary time 1016 ± 305 min, and MVPA time 83 ± 45 min) were included. For this reason, a pilot scheme based on an information campaign and the location of a specific brown container for biowaste in specific zones of the city was carried out in Castelló de la Plana (Spain) over a period of six months. In this period, the collection and composition of the biowaste was monitored in depth with the goal of determining the evolution of the efficiency of the new collection system over time. In the zones, the quality rate in the biowaste container increased as the pilot study progressed, finally reaching 90%. The rate of biowaste separation also increased in the three zones over time, although in different ways, which means that there is greater collaboration on the part of citizens. On the other hand, an analysis of the rate of net biowaste daily collection from zones 2 and 3 has shown that their value increases as the rate of containerization of biowaste decreases. Therefore, to obtain better results it will be necessary to increase the containerization of biowaste, that is, to reduce the distance from the citizen to the container. However, the mechanisms and criteria by which internalized CARDS toxin is transported and activated to execute its cytotoxic effects remain unknown. In this study, we used full-length CARDS toxin and its mutant and truncated derivatives to analyze how pharmacological drugs that alter pH of intracellular vesicles and electrical potential across vesicular membranes affect translocation of CARDS toxin in mammalian cells.
