In the present study we have tried to explore the effect of static external electric field of strength 3.0 V/nm on the conformational changes adopted by the protein β-lactoglobulin. We have chosen different temperatures viz. 300 K, 400 K and 450 K to evaluate the temperature dependent effect of electric field. We have observed that combined effect of high temperature and static external electric field show significant changes on the structural conformation of the protein which in turn may affect the functional properties of the protein. Calculations of root mean square deviations reveal that both helical and β-sheet regions of the protein are noticeably affected at high temperature. We have used solvent accessible surface area (SASA) and dipole moment values to explain that there is changes in hydrophobicity of the protein surface due to presence of external electric field. The study reveals that electric field in combination with high temperature can be used to alter the conformation of the protein and the effect of external electric field is more pronounced at high temperature than that of low temperature. The study provides a better understanding of the conformational changes adopted by the protein under the stress of external electric field and high temperature and provide guidance to choose optimum conditions for processing without loss of nutritional properties. The mammalian two-pore channels TPC1 and TPC2 are patho-physiologically relevant endo-lysosomal cation channels regulated by the Ca2+ mobilising messenger NAADP and the phosphoinositide PI(3,5)P2. Recent work by Arlt et al shows that genetic or chemical inhibition of TPC1 in mice promotes anaphylaxis in vivo through a mechanism involving enhanced endoplasmic reticulum Ca2+ release and secretion in mast cells.
Non-alcoholic fatty liver disease (NAFLD) is currently one of the most common chronic liver diseases especially in developed countries. Modern research shows an obvious protective effect of Sagittaria sagittifolia L. (Alismataceae) on glucose and lipid metabolism disorders. Previous studies had reported that Sagittaria sagittifolia polysaccharide (SSP) has potent protective effects on drug-induced liver injury. Based on this, we speculated that Sagittaria sagittifolia polysaccharide also has protective effects on NAFLD and performed experiments to explore this more.
Outstanding protective effects of SSP against NAFLD in mice was observed with Hematoxylin and Eosin (H&E) and uranium acetate-citrate stain in our prophase research. By performing bioinformatics analysis on plasma metabolic data which is obtained from ultra-performance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS), we found the regulatory mechanisms and key nodes behind the beneficial effect with IPA (Ingenuity Pathwa/HO-1 signaling pathway in liver oxidative stress, providing an attractive point for the breakthrough of related natural medicine development.
Immunosuppressive and immunomodulatory activity of mesenchymal stem cells derived from different sources, such as placental membranes, umbilical cord, and amniotic fluid has been proved. The heterogeneous nature of human amniocytes have been confirmed due to different clonal subpopulations found in amniotic fluid. The aim of this study was to investigate a 17-gene panel of immunomodulatory markers in two clonal subpopulations of CD90
amniocytes, divided based on morphology into epithelioid and fibroblastoid cells.
Semi-quantitative RT-PCR was used to study the expression of the chosen genes. Flow cytometry analysis confirmed the non-hematopoietic mesenchymal origin of isolated cells, based on lacking the hematopoietic marker of CD31, and the presence of mesenchymal marker of CD90 (both on more than 90% of cells).
Our results showed that besides growth characteristics, the two cell groups were different in expressional profile, so that, fibroblastoid clones displayed higher level of immunosuppression gnd high immunomodulatory potential, can help to understand the details of this relationship.Spiral artery (SpA) remodelling is essential for a successful pregnancy and is best described by its morphological features; vascular smooth muscle cell separation and loss, vessel dilatation, and invasion by extravillous trophoblast cells (EVT). Current opinion holds that EVT fully replace the endothelial cells (EC) of the SpA and take on EC-like characteristics. Placental bed biopsies (6-20 weeks gestation) were immunostained for EC and EVT, showing transient loss of EC. In vessels containing an endovascular EVT plug (n = 28) 77.6 ± 19.3% of the lumen was covered by EC while in vessels without endovascular EVT (n = 100) it was 100 ± 0%.The migration of the uranium (U) in high-level radioactive waste that is held in deep geological repositories via fractures in deep granite aquifers is a serious safety concern, thus, this study investigates the effect of fracture filling materials designed to mitigate these concerns. Geochemical analysis was conducted on granite rock core and groundwater samples collected from boreholes located in granite areas. Sequential extraction tests on fracture filling material (FFM) samples were also conducted. The rock core samples were classified as two-mica granite that had uranium (U) content ranging from 1900 to 22,100 μg/kg with an arithmetic mean of 8500 μg/kg. selleckchem The total U concentration in the FFM samples was found to range from 790 to 80,781 μg/kg. The U in the FFM samples was mainly associated with a carbonate phase that made up from 29.9 to 100% of the total U in the FFM. The U fraction of carbonate phase was closely correlated with the Ca fraction. U associated with crystalline inorganic FFM constituents (e.g, clay minerals and metal oxyhydroxides) was also found in FFM samples in fractions ranging from 21.1 to 70.1%. U in FFM is mainly incorporated via Ca-carbonate, which might have not been formed in modern groundwater, but the time and temperature during formation are unknown. In addition, the Fe, Si, Al, Ca, K, and U levels were found to be well correlated with each other, suggesting that U can also become geochemically associated with crystalline clay minerals or Fe-oxyhydroxides.
