Some AT4 ligand peptides, such as Ang IV and LVV-hemorphin-7, have been shown to act as IRAP inhibitors that exert memory-enhancing properties. As such IRAP has been a target for developing cognitive enhancers. To facilitate detailed mechanistic studies of IRAP catalysis and inhibition, and to pave the way for biophysical and structural studies of IRAP in complex with peptide inhibitors, we report here an optimized expression and purification system using High Five insect cells. We also report biochemical characterizations of the purified recombinant IRAP with a standard aminopeptidase substrate and an optimized IRAP peptide inhibitor with a Ki of 98 nM.Acute myocardial infarction (AMI) is the leading cause of death, morbidity, and health costs worldwide. In AMI, a sudden blockage of blood flow causes myocardial ischemia and cell death. Reperfusion after ischemia has paradoxical effects and may exacerbate the myocardial injury, a process known as ischemic reperfusion injury. In this work we evaluated the lipidome of isolated rat hearts, maintained in controlled perfusion (CT), undergoing global ischemia (ISC) or ischemia followed by reperfusion (IR). 153 polar lipid levels were significantly different between conditions. 48 features had q less then 0.001 and included 8 phosphatidylcholines and 4 lysophospholipids, which were lower in ISC compared to CT, and even lower in the IR group, suggesting that IR induces more profound changes than ISC. We observed that the levels of 16 alkyl acyl phospholipids were significantly altered during ISC and IR. Overall, these data indicate that myocardial lipid remodelling and possibly damage occurs to a greater extent during reperfusion. The adaptation of cardiac lipidome during ISC and IR described is consistent with the presence of oxidative damage and may reflect the impact of AMI on the lipidome at the cellular level and provide new insights into the role of lipids in the pathophysiology of acute myocardial ischemia/reperfusion injury.

    Central nervous system (CNS) tuberculosis (TB) is a calamitous infection with high rates of morbidity and mortality. Underlying HIV infection often increases susceptibility for acquiring TB and also complicates TB treatment. The study objectives were to assess the burden of CNS TB and associated factors in treatment experienced HIV infected adults.

    A single-center observational cross-sectional study was conducted between December 2019 and June 2020. Both descriptive and analytical statistics were used to analyze the data.

    Ninety-five HIV infected adults with presumptive TB-HIV co-infection on combination antiretroviral therapy for median of 144months were assessed. The mean age was 40.8years (1SD=12.4). Male to female ratio was 12. The prevalence of CNS tuberculosis was 56.8% (TB menigitis 53.7%, tuberculoma 2.1%, and spinal TB 1.1%). Patients with CNS TB were younger compared to those with extra CNS TB (38.6 vs. 43.6years, p=0.04).     We examined quality of education, literacy, and years of education in relation to late-life cognitive function and decline in older Puerto Ricans.

    Our sample consisted of 3,385 community-dwelling adults aged 60 years and older from the Puerto Rican Elderly Health Conditions study. Quality of education was based on principal component analysis of variables gathered from Department of Education and Census reports. Literacy (yes/no) and years of education were self-reported. Cognitive function was assessed in participants’ homes at baseline and 4 years later using a previously validated Spanish-language 20-point global screening measure for dementia, the minimental Cabán. Regression models were adjusted for sociodemographic and life course covariates.

    Quality of education was positively correlated with both educational attainment and cognitive performance. Independent of years of education, literacy, childhood economic hardship, and adult economic hardship, compared to participants in the lowest quartile of education quality, those in the highest quartile had significantly better baseline cognitive performance (
